清华大学果蝇中心 中 EN 主页 服务 Transgenic RNAi & Transcriptional activation (FlySAM) Microinjection CRISPR/Cas9 搜索 订购指南 常见问题 联系我们 lab members ... ... ... / 服务 / Transgenic RNAi Microinjection CRISPR/Cas9 果蝇的大部分基因与人类基因同源,并且很多与人类疾病相关的基因都与果蝇的相关基因同源。为了更好地研究这些基因的功能,转基因RNAi技术作为目前最便捷的一个研究工具,为筛选疾病相关的信号通路中新的调控因子提供了一个理想的平台。为了提高基因敲低效率,同时降低位置效应和脱靶概率,我们连续开发了第二、三代和新一代转基因RNAi技术。这些技术突破性地实现了果蝇生殖细胞中的RNAi,并且效率堪比体细胞RNAi (Ni et al., 2008, 2011, Nature Methods),而且可以实现对多个基因同时调控(Qiao et al., 2018, Nature Communications)。清华大学果蝇中心(THFC)成立于2011年,是为果蝇社区提供资源和技术服务的非营利性机构。目前,我们库存了大量转基因系果蝇以供生命科学和生物医学研究使用。 除了积累转基因RNAi资源,我们也针对果蝇开发了一套CRISPR/Cas9基因编辑系统 (Ren et al., 2013, PNAS),主要流程包括将编码sgRNA的质粒注射到在生殖细胞中特异表达Cas9的果蝇胚胎中,这个系统的主要特点是可遗传效率高且便于操作。 在基因的过表达(激活)方面我们基于CRISPR/Cas9开发了转基因激活技术,即flySAM(Jia et al.,2018, PNAS),该技术可以有效地激活一个或多个基因。 如果您对转基因RNAi品系、基因组定点编辑和转录激活感兴趣的话,请联系我们。同时,我们也对外提供载体构建和显微注射服务。 / 导航 / 搜索 订购指南 联系我们 清华大学 清华大学生命科学学院 清华大学医学院 电话: +86-010-62773106 邮箱: lpliu@mail.ts...
