Ela é causada por vírus, transmitido pelo mosquito da espécie haemagogus janthinomys, onde os macacos são os hospedeiros naturais. No entanto, os primatas não transmitem a febre amarela, somente pela picada do mosquito, nas áreas rurais. Já nas áreas urbanas, o Aedes Aegypti é o principal transmissor e reservatório do vírus que causa a Febre Amarela Urbana (FAU).
